In an end-to-end match, Pensilva belied their lowly League status and threw down the gauntlet to Looe, displaying some spirited attacking football.
However, it was Looe who took the lead when Matt Barton's cross was hammered home by Alec Cannon. With Pensilva still reeling, Looe went further ahead through Barton again.
Substitute Robbie Spencer made it three just after the break and the game looked over.
But the Pens did not quit and pulled one back.
With midfielder Danny Carroll off the field concussed, the home defence came under heavy pressure – and it was no surprise when they conceded another.
Spencer, found Barton, who made the score 4-2 before Spencer then scored his second and Looe's fifth.
Pensilva again reduced the arrears, and it was left to Spencer to banish any hopes of a visitors' comeback as he completed his treble to send his side through to the next round.
Looe deserved the win, just, but if Pensilva play like this for the rest of the season, their fortunes can only improve.
